Pyrano[3,2-c]quinolone structural motifs are commonly found in natural products with diverse biological activities. As part of a research programme aimed at developing the efficient synthesis of natural product-like small molecules, we designed and developed the microwave assisted, facile stereoselective synthesis of two series of carbohydrate fused pyrano[3,2-c]quinolone derivatives (n = 23) starting from 2-C-formyl galactal and 2-C-formyl glucal, reacting with various 4-hydroxyquinolones in shorter reaction times (15-20 min). The antiproliferative activity of these synthesized pyrano[3,2-c]quinolones was determined against MCF-7 (breast) and HepG2 (liver) cancer cells. The selected library members displayed low micromolar (3.53-9.68 μM) and selective antiproliferative activity. These findings on carbohydrate fused pyrano[3,2-c]quinolone derivatives are expected to provide new leads for anticancer drug discovery. © 2018 The Royal Society of Chemistry.

Stereoselective synthesis of natural product inspired carbohydrate fused pyrano[3,2-: C] quinolones as antiproliferative agents

We give a different formulation for describing maximal surfaces in Lorentz-Minkowski space, L3, using the identification of L3 with C × R. Further we give a different proof for the singular Björling problem for the case of closed real analytic null curve. Existence of maximal surface containing given curve and special singularity

Presence of cyclobutane rings in bioactive natural products makes them a popular synthetic target. Most common strategy for synthesizing cyclobutanes is [2+2] cycloaddition, which is usually facilitated by using ultraviolet radiation and catalysts. Herein we report the design and synthesis of densely functionalized cyclobutanes by alleviating these drawbacks and using sunlight. Further, we identified the putative mechanism of the transformation based on kinetic, fluorescence and crystallographic studies and evaluated biological activities of the products against MCF7 cell lines. The rational design of the olefinic substrates was based on their UV–vis spectra. The generic aspect of the reaction was elucidated by the syntheses of diverse analogs of cyclobutanes. Further, crystal structures and UV–vis spectra of the olefinic partners assisted us in rationalizing the stereoselectivity of the heterodimers. We believe this energy-efficient mild approach will provide a substantial contribution to the existing repertoire of strategies to regio- and stereoselectively access cyclobutanes. © 2018 Elsevier Ltd

<p>A facile cobalt(III) catalyzed C–H activation of arenes with substituted (benz)imidazoles as directing groups (DG) is reported. The strategy was utilized for the synthesis of polyheteroarenes when appropriate substrates were treated with diarylacetylenes as the coupling partner. The desired compounds were synthesized in moderate to excellent yield. A putative reaction mechanism is proposed. (Benz)Imidazole-Directed Cobalt(III)-Catalyzed C–H Activation of Arenes: A Facile Strategy to Access Polyheteroarenes by Oxidative Annulation

Elastic fibers are present in a variety of tissues and are responsible for their resilience. Until now, no optical contrast agent in the near-infrared (NIR) wavelength range of 700–900 nm has been reported for the imaging of elastic fibers. Here, we report the discovery of a NIR zwitterionic elastin probe ElaNIR (elastin NIR) through fluorescent-image-based screening. The probe was successfully applied for in vitro, ex vivo, and in vivo imaging by various imaging modalities. Age-related elastin differences shown by in vivo fluorescent and photoacoustic imaging indicated that ElaNIR can be a potentially convenient tool for uncovering changes of elastin in live models. Technological advancement of microscopic imaging has spurred the development of molecular probes for the detection of biologically and environmentally important analytes. Among fluorescent imaging tools, near-infrared (NIR) fluorescent molecules have been popular for in vivo imaging because of their ability to penetrate tissue deeper with reduced scattering and autofluorescence. Here, we report the discovery of the NIR zwitterionic elastin probe ElaNIR. We found that ElaNIR showed selective binding for elastin, such as those in the aorta, lungs, and skin. ElaNIR also allowed visualization and quantification of elastin levels in young and old mice, which reflect age-dependent changes of elastin in vivo. This makes ElaNIR a unique chemical tool for monitoring changes of elastin in tissue or animal models and provides new opportunities to study the effects of chemical compounds on elastin levels in biological processes. The near-infrared zwitterionic elastin probe ElaNIR for in vivo imaging of elastin structures was found through fluorescent-image-based screening. ElaNIR is well suited for in vitro, ex vivo, and in vivo imaging of elastin and also allows visualization and quantification of elastin levels in young and old mice, which reflect age-dependent changes of elastin in vivo. © 2018 Elsevier Inc.

Seeing Elastin: A Near-Infrared Zwitterionic Fluorescent Probe for In Vivo Elastin Imaging

Two polymers based on (3E,7E)-3,7-bis(2-oxoindolin-3-ylidene)benzo[1,2-b:4,5-b′]difuran-2,6(3H,7H)-dione (BIBDF) coupled with (E)-2-(2-(thiophen-2-yl)vinyl)thiophene (TVT) or dithienylbenzothiadiazole (TBT), namely PBIBDF-TVT and PBIBDF-TBT were synthesized via the Stille coupling reaction. The effect of benzothiadiazole or vinylene-π spacer of the copolymers on optical properties, energy levels, electronic device performance and microstructure were studied. It was found that PBIBDF-TBT based OFET devices, annealed at 180 °C, showed better performance with the highest electron mobility of 2.9 × 10-2 cm2 V s-1 whereas PBIBDF-TVT polymer exhibited 5.0 × 10-4 cm2 V s-1. The two orders of magnitude higher electron mobility of PBIBDF-TBT over PBIBDT-TVT is a clear indicator of the better charge transport ability of this polymer semiconductor arising from its higher crystallinity and better donor-acceptor interaction. © 2018 The Royal Society of Chemistry.

Porous organic materials are an emerging class of functional nanostructures with unprecedented properties. Dynamic covalent assembly of small organic building blocks under thermodynamic control is utilized for the intriguingly simple formation of complex molecular architectures in one-pot procedures. In this Review, we aim to analyze the basic design principles that govern the formation of either covalent organic frameworks as crystalline porous polymers or covalent organic cage compounds as shape-persistent molecular objects. Common synthetic procedures and characterization techniques will be discussed as well as more advanced strategies such as postsynthetic modification or self-sorting. When appropriate, comparisons are drawn between polymeric frameworks and discrete organic cages in terms of their underlying properties. Furthermore, we highlight the potential of these materials for applications ranging from gas storage to catalysis and organic electronics.
